Brief Introduction:
Ondansetron is a 5-HT3 receptor antagonist primarily used as an antiemetic. It has potential therapeutic applications for neuropathic pain, but its permeability into the nervous system may be limited by P-glycoprotein (Pgp) at the blood-brain barrier. In a study involving rats, a validated LC-MS/MS method was used to measure ondansetron concentrations in serum and brain microdialysates, demonstrating the drug's pharmacokinetics in both matrices. This method was highly sensitive and required only small sample volumes, enabling frequent sampling and a comprehensive pharmacokinetic profile. Additionally, a retrospective study in emergency department patients found that ondansetron administration was not associated with a significantly increased risk of dysrhythmias, including torsades de pointes, even in patients with a QTc interval ≥500 ms, suggesting that it may be safe to administer ondansetron to patients with prolonged QTc intervals.
